摘要
目的 建立一种测定血浆中头孢他美浓度的方法。方法 以HypersilBDSC18柱作分析柱 ,流动相为甲醇 -水 -冰醋酸 (2 8∶72∶1) ,流速为 0 .8ml·min-1,检测波长 2 6 2nm。结果 血浆中头孢他美的最低检出浓度为0 .16 μg·ml-1,3种浓度回收率为 99.2 %~ 10 0 .0 % ,日内 RSD ≤ 2 .5 7% ,日间 RSD ≤ 4.6 9%。结论 该法简便、灵敏、准确可靠 。
OBJECTIVE A high-performance liquid chromatographic method was developed for determination of cefetamet in dog plasma. METHODS The plasma samples were directly analyzed using BDS C 18 column after protein precipitation with methanol and being centrifuged at a speed of 10000 r·min -1. The mobile phase was methand-water-glacial acetic acid =28∶72∶1, the flow rate was 0.8 ml·ml -1 and the eluate was monitored at 262 nm. RESULTS The detection limit of cefetamet in plasma is 0.16 μg·ml -1, the recovery range of three different concentration samples was 99.2%~100.0%, the RSD within-day was no more than 2.57% and the RSD between-day was no more than 4.69%. CONCLUSION The method was simple?sensitive and accurate,and may be applied to the pharmacokinetic study of cefetamet in vivo.
