摘要 目的 :研究睫状神经营养因子 (CNTF)对脊髓损伤后逆行性红核神经元损伤的保护作用和促进肢体功能恢复的作用。方法 :将大鼠分为假手术组、生理盐水 (NS)组和 CNTF组 ,用 Allen改良法撞击致 SD大鼠脊髓 T1 0 损伤 ,经蛛网膜下腔留置导管注射 NS和 CNTF;术后每周进行一次 BBB评分和斜板试验的行为学评分 ;6周时采用辣根过氧化物酶 (HRP)逆行示踪技术标记红核神经元 ,取红核部位脑组织作冰冻切片 ,应用四甲基联苯胺 (TMB)呈色反应显示脊髓损伤后红核神经元存活率 ,并进行图像数据的分析。 结果 :(1) CNTF组行为功能评分高于 NS组 ;(2 ) CNTF组 HRP标记红核神经元数目多于对照组。 结论 :CNTF可减轻脊髓损伤后的逆行性损伤 ,而对上行运动神经元发挥保护作用 ,从而对肢体功能的恢复起到促进作用。 Objective:To study the neuroprotective effects of ciliary neurotrophic factor(CNTF) on rubrospinal neurons and functional recovery in a contusion model of spinal cord injury (SCI) in the adult rat. Methods:SD rats were divided into 3 groups:sham operated group, NS and CNTF group. NS and CNTF were given through intrathecal catheter for 10 d after T 10 were injured by modified Allen contusion method. Animals were behaviorally tested for 6 weeks using the Basso, Beattie, Bresnahan locomotor rating scale and inclined plane test. At the end week 6, rubrospinal neurons of 5 rats in each group were labeled by retrograde transport of the horseradish peroxidase (HRP) from the lesion site, which were taken by damaged axons and remained in the neurons, then the labeled red nucleus neuron (RN) were counted. Results:(1) Rats treated with CNTF had more improvements in hindlimb function than controls. (2) The amount of survival rubrospinal neurons was significantly higher in CNTF treated animals than in controls. Conclusion:Intrathecal infusion of exogenous CNTF following SCI significantly reduces retrograde neuronal cell death and enhances functional recovery. CNTF plays a neuroprotective role against spinal cord injury in the adult rat.
