TGF-βⅡ型受体mRNA及蛋白在人IgA肾病肾小管上皮的表达 被引量:1

摘要 为探讨转化生长因子 -β (TGF-β)在人 Ig A肾病肾小管间质纤维化中的作用 ,本文用原位杂交及免疫组织化学方法研究转化生长因子 -β 型受体 (TGF-βR ) m RNA和蛋白在肾小管上皮细胞的表达。结果表明 :TGF-βR m RNA定位于肾小管及集合管上皮细胞。 TGF- βR 蛋白定位于肾小管及集合管上皮细胞的基底面、腔面及侧面 ,或呈与基底膜相垂直的基底部纵纹出现于一些细胞的胞质内。肾小球系膜细胞也可见 TGF- βR m RNA及蛋白表达 ,但不如肾小管明显。以上结果提示肾小管和集合管上皮细胞的受体可与 TGF- β特异性结合。 TGF- β则通过自分泌作用促进小管上皮细胞合成与分泌ECM。过多的 ECM在小管周围沉积 ;或 TGF-β抑制 ECM的降解 ,终将导致 Ig In order to investigate the effect of transforming growth factor β(TGF β) on the interstitial fibrosis in human IgA nephropathy, TGF β type Ⅱ receptor (TGF βRⅡ) expression was studied with in situ hybridization and immunohistochemistry. The results showed that: TGF βRⅡ mRNA localized in the epithelial cells of renal tubules and collecting tubules. The TGF βRⅡ protein expressed at basal, free and lateral surface of epithelial cells of renal tubules and collecting tubules. Within some spithelial cells the positive reaction was demonstrated in basal striations arranged in a vertical orientation to basement membrane. The expressions of TGF βRⅡ mRNA and its protein in the mesangial cells of glomerulus were less obvious than that in epithelial cells. It was suggested that TGF β binding to TGF βRⅡ in epithelial cells may play a key role in promoting the cells of tubules to synthesige and secrete ECM by may of autocrine. The excessive accumulation of ECM around tubules will result in interstitial fibrosis in IgA nephropathy.\;
