摘要
目的在骨缺损中植入骨形成细胞和藻酸盐复合物,观察缺损的修复情况。方法体外扩增兔骨髓基质干细胞(bonemarrowstemcell,MSCs),将MSCs与2.0%藻酸钠溶液混合形成MSCs/藻酸钠复合物,使细胞的终浓度为5×106/ml。将MSCs/藻酸钠复合物用氯化钙固化为凝胶体,植入细胞供体兔颅骨直径15mm缺损中,以单纯植入藻酸盐凝胶作为对照。术后8周取材,通过大体观察、X线检查、组织学检查观察骨缺损的修复情况。结果MSCs/藻酸钠凝胶植入8周后,缺损大部分获得修复,组织学分析显示缺损中有大量新骨形成。结论应用藻酸钠复合MSCs可以提高骨缺损修复效果。
Objective The study investigates the utilization of alginate carrier for delivering rabbit marrow stem cells(MSCs )and creating new bone tissue in the repair of bone d efects.Methods Rabbit MSCs cells were cultured and e xpanded in vitro.The MSCs were mixed with 2.0%alginate sodium solution to generate MSCs /alginate composites with fin al cellular density of 5×10 6 /ml.Calcium chloride was used as cross-linking agent to gel aqueous a lginate solution.The MSCs /alginate composites /alginate composites were implanted into rabbit skull defe cts in the diameter of 15mm.Eight weeks a fter operation,the animals were sac rificed and new bone regeneration in the defects was investigated by gross specimen inspection,X-ray ex amination and histological observation.Results8weeks after operation,most area of the MSCs /alginate composites transplanted defects we re filled with newly formed bone.Conclusion These results revealed that MSCs /alginate composites may be feasible in the repair of bone defects.
