摘要
目的 :从山羊胎皮提纯羊Ⅲ型前胶原 ,以代替人Ⅲ型前胶原生产免疫试剂。方法 :以山羊胎皮为原料 ,通过 11 2 %硫酸铵及 10 %氯化钠盐析和超速离心 ,DE32和DE5 2阴离子交换纤维素柱层析提取纯化得到羊Ⅲ型前胶原。结果 :测得羊Ⅲ型前胶原分子量为 4 6 8kDa。主要成分甘氨酸、羟脯氨酸和脯氨酸构成比分别为 4 0 4 %、11 7%和 11 1%。与人羊Ⅲ型前胶原的交叉反应为 6 5 89%。结论
Aims:Goat procollagen type Ⅲ(gPCⅢ)was isolated and purified from their fetal skin instead of immuno-reagent of human PCⅢ.Methods:Purified gPCⅢ was acquired from the material of goat fetal skin through salting out(11.2% ammonium sulfate and 10% sodium chloride),ultracentrifuge and ion-exchange cellulose chromatography(DE32 and DE52).Results:Molecular weight of gPCⅢ determined was 468 kDa.The ratios of main components of glycine,hydroxyproline and proline were 40.4%,11.7 and 11.1% respectively,The cross reaction with human PCⅢ was 65.89%.Conclusion:The isolated gPCⅢ was acquired in this method may be instead of immuno-reagent of human PCⅢ.
