摘要
目的 观察烧伤小鼠 2 4h内肝组织糖皮质激素受体在蛋白及mRNA水平的时相变化特点。方法 将 6 0只BALB/C小鼠随机分为正常对照组和实验组 ,实验组给予背部Ⅲ度 15 %~ 2 0 %烧伤。分别于伤后 2、4、6、12、2 4h测定血清皮质酮浓度 ,用Westernblotting、RT -PCR在蛋白质和mRNA水平测定肝组织糖皮质激素受体。结果 烧伤小鼠血清皮质酮浓度 2~ 4h达到高峰 ,2 4h仍未恢复正常 ;肝组织糖皮质激素受体在蛋白质水平 4~ 6h降至最低 ;而mRNA水平仅仅在伤后 2h有短暂降低。结论 烧伤应激引起小鼠血清皮质酮水平明显升高 ,其受体在蛋白及mRNA水平均可降低 ,但蛋白的降低不仅仅发生在mRNA水平。
Objective To observe the changes of hepatic glucocorticoid receptor(GR)at protein and mRNA level in burned mice.Methods 60 BALB/C mice were divided randomly into control and burn group.After clipping of the hair,15%~20% thickness third degree burn injury was produced.At 2,4,6,12,24 h after burn,serum corticosterone concentration was examined,hepatic GR and GR mRNA were detected by Westernboltting and RT-PCR.Results After burn injury,serum corticosterone level was markedly elevated at 2~4 h and hepatic GR was markedly downregulated at 4~6 h.The reduced GR mRNA was only observed at 2 h.Conclusions Serum corticosterone level was markedly elevated and hepatic GR was downregulated at protein and mRNA level.It is not principal machanism about downregulation of GR at mRNA lever.
