摘要
目的 初探KAI1/CD82在肠癌组织中的表达水平及可能涉及的机制。方法 采用多重逆转录聚合酶链反应 ,对 18例肠癌组织和正常肠组织中KAI1/CD 82mRNA表达水平进行半定量检测。用去甲基化试剂 5 氮脱氧胞苷作用于Bel 740 2和SMMC 772 1肝癌细胞株 ,然后检测KAI1/CD82mRNA表达水平。结果 肠癌组织中KAI1/CD82mRNA表达水平 (0 .78±0 .2 2 )较正常肠组织中表达水平 (1.5 3± 0 .12 )显著降低 (P <0 .0 5 ) ,其中 5例肠癌患者有转移 ,癌组织中KAI1/CD82mRNA表达水平 (0 .70± 0 .2 4) ,与未转移组没有显著差异。 5 氮脱氧胞苷作用下 ,SMMC 772 1细胞株中KAI1/CD82mRNA表达水平基本没变 ,而Bel 740 2细胞株KAI1/CD82mRNA表达水平有一定的增高。结论 肠癌组织中KAI1/CD82mRNA表达水平较正常肠组织中表达显著降低 ,这种表达降低可能涉及到甲基化等多种复杂调控的机制。
Objective To investigate the expression levels of KAI1/CD82 mRNA in intestinal tumour and normal intestinal tissues and the regulative mechanism of KAI1/CD82 expression. Methods KAI1 mRNA of 18 cases with intestinal tumour as well as, normal intestinal tissues were detected using duplex reverse transcription polymerase chair reaction (duplex RT-PCR)method.Bel-7402 and SMMC-7721 HCC cell lines were cultured after being demethylated by 5-aza-deoxycytidine.Results The average expression levels of KAI1/CD82 mRNA in intestinal tumour tissues(0.78±0.22)were significantly by lower than that in normal intestinal tissues(1.53±0.12)P<0.05 .KAI1/CD82 expression of intestinal tumour metastasic group(0.70±0.24)weren't significantly lower than that of non-metastasic group.After being demethylated by 5-aza-deoxycytidine,the expression level of KAI1/CD82 mRNA in Bel-7402 cell increased,but could't detect significant change in SMMC-7721 cell.Conclusion The indicated KAI1/CD82 in intestinal tumor might be a marker for intestinal tumour cell migration and that several regulatory mechanisms including gene demethylation might be involved in KAI1/CD82 expression.
