摘要
目的 :建立反相高效液相色谱法 (HPLC)测定犬血清中加替沙星浓度。方法 :血清样品用二氯甲烷提取 ,以环丙沙星为内标 ,流动相为甲醇 - 0 .0 1 mol/L磷酸二氢钾缓冲液 - 0 .5mol/L四丁基氢氧化铵溶液 (1 5∶ 75∶ 1 .4) ,磷酸调p H至 2 .6;流速为 1 .0 mg L /min;检测波长 2 98nm。结果 :本法在 0 .2~ 8.1 1μg/ml范围内线性良好 ,r=0 .9998,日内RSD为 3.9%~ 7.3% ,日间 RSD为 5.7%~ 7.3% (n=5) ;血清最低检测浓度为 0 .0 5μg/ml。结论 :本法快速、准确 ,重现性好 。
Objective:To establish a HPLC assay method for the determination of Gatifloxacin in dog serum.Methods:A reversed phase ion Pair chromatographic method was developed after the serum samples were extracted by dichloromethane solvent.HPLC conditions were set as follows:Column:Li Chrosorb C 18 ,250mm×4.6mm,10μm;Mobile phase methanol-0.01mol/L potassium dihydrogen phosphate 0.5mol/L tetra butyl ammonium hyeroxygen(15:75:1.4),pH was adjusted to 2.6 with phosphoric acid;Flow rote:1.0ml/min,Detection wave length:298nm.Results:Assay linearity was obtained in the range of 0.20~8.11μg/ml; Intra day and inter day RSD were 3.9%~7.3% and 5.7~7.3% ( n =5),respectively.The lowest detectable concentration was 0.05μg/ml.Conclusion:The method is rapid,accurate and reproducible.It is suitable for therapeutic drug monitoring(TDM).
