摘要
以玉米自交系 792 2的幼胚为外植体 ,全面探讨了影响其愈伤组织诱导的主要因素 ,如接种方式、4℃保存时间、碳源种类及其浓度、培养基成分、AgNO3等。结果表明 :采用盾片侧向上的方式 ,取 4℃保存时间不超过 7天的幼胚 ,以 8114、MB、MS、N6为基本培养基 ,蔗糖浓度为 4 0g/L或用 2 0g/L的蔗糖 + 10g/L的葡萄糖为较适宜的诱导条件。
Factors influencing the callus induction of immatue embryos in maize inbred line 7922 such as ways of inoculation of immature embryos,duration of 4℃treatment,culture medium,carbon source and their concentration,were studied.The results showed that the better conditions of callus induction were the way of inoclation using scutella up,dureation of 4℃ treatment being lower than 7 days,8114,MB,MS,N6 being used as culture medium,concentration of sucrose being 4 g/L or 20 g/L glucose.The effect of AgNO 3 was also studied.The result showed that AgNO 3 had positive effect on callus induction.
