摘要
目的 研究表明丝氨酸蛋白酶及其抑制剂参与脑缺血引起的神经元损伤。本研究初步观察糜蛋白酶样丝氨酸蛋白酶抑制剂 N-甲苯磺基 - L-苯乙胺酰氯甲基酮 (N- tosyl- L- phenylanyl chloromethyl ketone,TPCK)对局灶性脑缺血的保护作用。方法 利用大鼠线栓法局灶性脑缺血模型 (脑缺血 1h/再灌注 2 3h)及激光多普勒血流计、氯化三苯基四氮唑 (2 ,3,5 - triphenyltetrazolium chloride,TTC)染色和图像分析仪 ,观察 TPCK对半暗带脑血流 (Penumbral cerebral blood flow,p CBF)和梗死体积的影响。结果 TPCK虽不影响 p CBF,但可缩小梗死体积。结论 糜蛋白酶样丝氨酸蛋白酶参与脑缺血继发性损伤 。
Objective It has been showed that the serine proteases and their inhibitors involve in the damage caused by cerebral ischemia.In the study,we observed the protection of TPCK,a chymotrypsin like inhibitor of serine proteases,against local cerebral ischemia preliminarily.Methods The rats were subjected to local cerebral ischemia lasting for about 1h and reperfusion for 23h.We measured the cerebral blood flow(CBF) in the penumbra by laser doppler flowmetry,and determined the infarct volume by TTC staining and image analysis.Results Although TPCK had no effect on the CBF in penumbra,it reduced the infarct volume caused by local cerebral ischemia (P<0.01).Conclusion The chymotrypsin like serine protease involves in the ischemic damage of brain and their mechanisms will be studied profoundly.
