摘要
为研究一种相对准确且简单的鸡精子密度的检测方法,对黑丝羽乌骨鸡及其杂交公鸡的精液,分别运用计数板法、吸光度(精子密度仪)法和视野精子数法进行精子密度的测定,分析计数板法测定的密度与吸光度、视野精子数间的相关性。结果表明,计数板法测定的密度与吸光度之间的回归关系为幂函数回归,回归关系极显著(P<0.01);计数板法测定的密度与视野精子数之间的回归关系为直线回归,回归关系极显著(P<0.01)。因此,吸光度法和视野精子数法均能准确地测定鸡精子密度,比计数板法简单。吸光度法利用建立的标准曲线能够快速、精确测定精子密度,但需要专用的分光光度计或者精子密度仪;视野法采用便携式带液晶屏的温控显微镜,能使计数更直观简便,能相对快速准确地测定精液密度,同时还能与精子活力评价相结合,具有很好的推广应用价值。
In order to explore an accurate and simple method to detect sperm density in chicken, semen was collected from different roosters of black silky chicken and its hybrids by massage. The regression and correlation analysis (RCA) between sperm concentration and sperm number in the field of microscope vision (SNMV), and RCA between sperm concentration and semen absorption spectrum (SAS)were studied. The results showed that RCA of sperm concentration and SAS was powerful function model, and there was extremely significant,correlation between RCA of sperm concentration and SAS(P〈0.01). The RCA of sperm concentration and SNMV was linear regression, and there was also extremely significant correlation between RCA of sperm concentration and SNMV (P〈0.01). Statistical analysis showed that both SAS and SNMV was fast and accurate for sperm density test. The SAS could be used to test the sperm density quickly and accurately by the standard curve established,but it needed special spectrophotometer or sperm density meter. It had become much more conveniently, quickly and accurately to test semen density using SNMV by portable LCD display as well as temperature control microscope. Especially,SNMV could also be used to test the sperm viability. So,SNMV was very useful to determine sperm quality both in field and in research in the future.
