摘要 目的 :构建粘细菌发育特异性转录因子FruA相关蛋白FruB的敲除菌株 ,初步阐明FruB在粘细菌发育周期中的作用。方法 :在fruB基因中央编码区插入四环素抗性基因 ,导入野生型粘细菌DZF1后通过双交叉同源重组途径阻断基因组内fruB的功能。结果 :抗性筛选和PCR检测显示分离出了fruB基因阻断菌株fruB ::TcΩ2。fruB功能缺失延缓子实体的发育及营养细胞向粘孢子分化。结论 :fruB与粘细菌发育分化存在相关性。 Objective FruB is an associated protein of FruA,a transcription factor essential for the development of Myxococcus xanthus .This work was performed to knock out the wild type allele of fruB and evaluate the developmental process of the mutant strain.Method DNA fragment conferring tetracycline resistance was inserted into the central coding region of fruB .The resulting plasmids were transferred into M.xanthus DZF1.Colonies were selected with double homologous cross over method.Results Based on antibiotic screening and PCR examination, fruB disruption mutants,designated fruB ::TcΩ2,were obtained.Characterization of fruB ::TcΩ2 indicated that disruption of fruB delayed the morphogenesis of fruiting bodies and depressed spore yield.Conclusion FruB may be involved,to some extent,in the development of M.xanthus.
