摘要 双向聚丙烯酰胺凝胶电泳按等电点和分子量对蛋白质进行分离,具有很高的分辨率,已经成为蛋白质组研究中的核心技术之一。目前的方法还难以满足真核生物蛋白质组研究的要求。自1975年O’Farrell提出该方法以来,针对应用中常见的一些问题,如样品的增溶及加载,样品的预分级分离,阴极漂移和凝胶上蛋白质的回收等,对这种技术进行了不断的改进。 Two-dimensional polyacrylamide gel electrophoresis(2D-PAGE)is a highly resolving technique for pro-teins separation by isoelectric point and molecular mass,which is being more and more critical as the fundamental separation method for proteome research.But currently available methods are not satisfied with the most eukaryotic proteomes.Since the description of the original technique by O'Farrell in1975,numerous studies have contributed to the improvement of the performance of2D-PAGE.Solutions to the problems most frequently encountered by2D-PAGE users are given constantly,such as the solubilization and loading of the sample,prefractionation of the sam-ple,cathodic drift and recovery of the proteins from the gel.These developments were described.
