木霉菌的形态学和可溶性蛋白质电泳鉴定与分类 被引量:7

摘要 对 6 1株来自不同植物根际的木霉菌 (Trichoderma spp.)进行了形态学鉴定 ,其中哈茨木霉菌 (Trichoderma harzianum)有 47株 ,绿色木霉菌 (Trichoderma viride) 1 0株 ,康宁木霉菌(Trichoderma koningii)和绿木霉菌 (Trichoderma virens)分别为两株。对这些菌株进行了可溶性蛋白质电泳分析 ,并采用 UPGMA法进行聚类分析。结果表明 ,将距离系数 D=1 3 .8作为聚类分析阈值 ,6 1个菌株可分为 A,B,C三个组 ;距离系数 D=9.1时 ,A组可分为 A1 ,A2 ,A3 ,A4,A5五个亚组。 A组包括 47株哈茨木霉菌 ,是主要成员。此外 A组还有 8株绿色木霉菌 ,两株绿木霉菌和一株康宁木霉菌 ,B组只有一株绿色木霉菌 ,C组由一株绿色木霉菌和一株康宁木霉菌组成 ,表明木霉菌具有明显的形态学多样性 ,而哈茨木霉菌则存在种下分类群 ,还发现蛋白质电泳分类结果与菌株的地缘有关。 Sixty one isolates of Trichoderma spp. isolated from different plant rhizosphere were identified morphologically as 47 Trichoderma harzianum, 10 Trichoderma viride, 2 Trichoderma koningii and 2 Trichoderma virens respectively. UPGMA dendrogram of all the isolates was produced by soluble protein gel electrophoresis(SPGE) data and showed 3 groups (A, B, and C) at the taxonomic distance index 13.8. Group A was divided into subgroups A1, A2, A3, A4, A5 at the distance 9.1, and included 47 Trichoderma harzianum which was the main members, 8 Trichoderma viride, 1 Trichoderma koningii and 2 Trichoderma virens. Group B included only 1 Trichoderma viride. Group C included 1 Trichoderma viride and 1 Trichoderma koningii. These results showed that Trichoderma spp. diversed morphologically and sub-taxonomic groups might exist in Trichoderma harzianum. The grouping obtained with SPEG method was found to be in accordance to geographic variation of the isolates.
