总抗氧化能力和一氧化氮合酶对大鼠隐睾生殖细胞凋亡的影响 被引量:9

The effect of total antioxident capacity,nitric oxide synthase on the germ cell apoptosis in the rat experimentally cryptorchid

摘要 目的 探讨总抗氧化能力 (T AOC)和一氧化氮 (NO)合酶 (NOS)对隐睾生殖细胞凋亡的影响。方法 SD雄性大鼠日龄 2 2d时复制单侧隐睾模型。用生物素 dUTP/酶标亲和素测定法检测睾丸生殖细胞凋亡 ;用硝酸还原酶法测定睾丸组织中NO含量 ;用化学比色法测定睾丸组织中T AOC、NOS活性。结果 术后第 7天 ,与对侧睾丸相比 ,隐睾发生凋亡的生殖细胞数显著增加 ,NO含量及NOS活性显著上升 (P <0 .0 1 ) ,在隐睾组织中两者呈正相关 (rs =0 .890 ,P <0 .0 1 ) ;T AOC显著降低 (P <0 .0 1 )。结论 隐睾组织中NO和NOS升高、T AOC下降是隐睾生殖细胞凋亡增加的生化机制之一。 Objective To study the effect of total antioxident capacity(T AOC) and nitric oxide synthase (NOS) on the germ cell apoptosis in the cryptorchid.Methods Immature rats (22 day old Sprague Dawley) were subjected to unilaterally cryptorchid.Germ cell apoptosis was detected by terminal deoxynucleotidyl transferase mediated d UTP nick end labeling (TUNEL).Biochemical parameters (T AOC,NO,NOS) were evaluated with spectrophotometric determination.Results At the 7th day after the operation,as compared to the control,the number of apoptotic germ cell in the cryptorchid was increased significantly,but its testes weight was decreased predominantly (P<0.01).The levels of NO and NOS in the cryptorchid were significantly higher than the control,both were positive correlation (r s=0.890,P<0.01),and the level of T AOC was predominantly lower (P<0.01).Conclusion The levels of T AOC,NO and NOS might be involved in the germ cell apoptosis in the cryptorchid.
