摘要
目的 探讨孤啡肽在急性吗啡耐受形成中的作用。方法 本实验采用热甩尾观察和反转录多聚酶链式反应(RT PCR)等方法 ,对大鼠急性吗啡耐受形成中痛行为 ,海马孤啡肽 (OFQ)mRNA表达进行检测 ,并通过侧脑室注射孤啡肽受体反义寡聚核苷酸 ,以抑制OFQ作用 ,来观察其对急性吗啡耐受形成的影响。结果 伴随急性吗啡耐受的形成 ,海马中OFQmRNA呈现升高趋势。预先以孤啡肽受体反义寡聚核苷酸处理 ,AMT形成受抑制。
Objective To study the effects of orphanin FQ(OFQ) on the development of acute morphine tolerance(AMT). Methods Wistar rats were given 5 consecutive subcutaneous injections of morphine hydrochloric acid (6mg/kg) to establish AMT rat model. The pain threshold (PT) was determined by hot water induced tail flick test and the expression of OFQ mRNA was measured by RT PCR. Results PT was significantly increased 1 h after 5 consecutive s.c MOR in rats, which were treated by i.c.v OFQ receptor antisense oligo deoxynucleotides (ODNs) in advance .The expression of OFQ mRNA was increased 1 h after 5 consecutive injections. Conclusion The increase of OFQ may be an important reason in the development of AMT.
