头痛安对偏头痛模型大鼠β-EP、CGRP、ET的影响 被引量:14

摘要 目的考察头痛安(由川芎、白芷、乳香、天麻、冰片等组成)对偏头痛模型大鼠的镇痛作用及机制。方法采用皮下注射硝酸甘油(GNT)诱发偏头痛大鼠模型,并予头痛安滴鼻干预,从行为学药效指标和β-内啡肽(β-EP)、降钙素基因相关肽(CGRP)、内皮素(ET)等生化因子两个方面观察头痛安对偏头痛模型大鼠的影响。结果对比空白对照组,模型组(GNT剂量为10 mg·kg^(-1))大鼠有明显的耳红、挠头、爬笼等偏头痛的行为学症状,同时脑组织中的β-EP降低,血浆中的CGRP、ET含量升高;对比模型组,阳性对照组(正天丸组)和头痛安各剂量组大鼠耳红和挠头的消失时间缩短,在30~180 min内各个时间段的挠头次数和爬笼次数减少,β-EP升高,而CGRP、ET含量降低;其中头痛安中剂量组、高剂量组上述指标改变明显,尤以高剂量头痛安效应最显著,类似于甚至优于正天丸。结论大鼠皮下注射GNT能有效诱发出偏头痛行为学症状,这种血管性偏头痛的发生可能与大鼠体内β-EP、CGRP、ET等的作用有关,高剂量头痛安可能通过调节这些血管活性物质和神经递质,抑制血管扩张和神经源性炎症反应而发挥镇痛作用。 Objective To investigate the effect and mechanism of Toutongan on migraine model rats. Methods Weuse the method of "subcutaneous injection of nitroglycerin(GNT) "to prepare the migraine headache model. Theeffect of Toutongan on praxiology,and biochemical factors such as β-EP,CGRP and ET of the model rats wereobserved. Results Compared with control group,behavioral symptoms of migraine such as red ear,head scratchingand cage climbing were obvious in model group rats,as well as reduced β-EP in brain tissue,increased CGRP andET in plasma. Compared to the model group,the duration of red ear and head scratching was shorten in positivecontrol group and all doses Toutongan groups;the counts of head scratching and cage climbing were less within 30-180 minutes, with the contents of β-EP increased, CGRP and ET decreased. The above indexes were changedsignificantly in middle dose and high dose Toutongan groups,specially,the analgesic effect was the most significantin high dose Toutongan group,which was similar to or even better than Zhengtian pill. Conclusion Subcutaneousinjection of nitroglycerin could effectively induce migraine behavioral symptoms;the occurrence of such vascularmigraine may be related to β-EP, CGRP and ET. High dose Toutongan could adjust these vascular activesubstances and neurotransmitters,and inhibit vasodilation and neurogenic inflammation to exert analgesic effect.
