肝激酶B1对肝癌细胞增殖、迁移的影响及其与患者术后预后的关系 被引量:4

Effect of liver kinase B1 on biological function of hepatocellular carcinoma cells and its relationship with postoperative prognosis

摘要 目的探讨肝激酶B1(LKB1)对肝癌细胞生物学功能的影响及与术后复发的关系。方法采用分子生物学技术构建LKB1过表达慢病毒载体,包装慢病毒感染高转移肝癌细胞MHCC97H,建立LKB1对照MHCC97H细胞株(对照组)和LKB1过表达MHCC97H细胞株(观察组)。采用细胞计数试剂盒(CCK-8)分析对照组和观察组细胞的增殖能力,采用Transwell分析对照组和观察组细胞的侵袭能力。并选取我院接受治疗的142例晚期肝癌患者作为研究对象,取肿瘤组织,免疫组织化学分析LKB1表达,并分组分析LKB1高表达和低表达患者术后复发率。结果成功构建LKB1过表达MHCC97H细胞株。与对照组细胞比较,观察组细胞株MHCC97H细胞的增殖能力明显下降,差异有统计学意义(P<0.05)。与对照组细胞[(87.12±9.25)个]比较,观察组细胞的侵袭能力[(28.32±7.32)个]显著下降,差异有统计学意义(P<0.05)。免疫组织化学分析LKB1低表达组患者98例,比率为69.01%,高表达44例,比率为30.99%。高表达组患者术后2年和3年复发率分别为27.27%和47.62%,低表达组患者术后2年和3年复发率分别为40.82%和68.37%,两组差异有统计学意义(Log-rank=2.013,P<0.05)。结论LKB1与肝癌细胞的增殖和迁移能力有关,低表达LKB1患者术后复发率明显增加。 Objective To investigate the effect of liver kinase B1 (LKB1) on the biological function of hepatocellular carcinoma cells and its relationship with prognosis.Methods LKB1 overexpression lentiviral vector was constructed by molecular biology technology, and the MHCC97H cells with high metastasis were infected by lentiviruses. LKB1 control MHCC97H cell lines (control group) and LKB1 overexpression MHCC97H cell lines (observation group) were established. The proliferation ability of cells was analyzed by cell counting kit-8 (CCK-8) assay in control group and observation group. The invasion ability of cells was analyzed by Transwell in control group and observation group. 142 patients with liver cancer from June 2012 to June 2018 in our hospital were selected as subjects. The expression of LKB1 was analyzed by immunohistochemistry and the recurrence rate of patients was analyzed in LKB1 high expression and low expression groups.Results The LKB1 overexpression MHCC97H cell line was successfully constructed in this study. As compared with control group, the proliferative capacity of MHCC97H cells in observation group was significantly decreased (P<0.05). As compared with control group, the invasive ability of cells in the observation group [(28.32±7.32) cells] was significantly decreased (P<0.05). Immunohistochemical analysis showed there were 98 cases (69.01%) in the LKB1 low expression group and 44 cases (30.99%) in the LKB1 high expression group. The 2- and 3-year recurrence rate was 27.27% and 47.62% in the high-expression group, and 40.82% and 68.37% in the low-expression group, respectively. There was significant difference between the two groups (Log-rank=2.013, P<0.05).Conclusion LKB1 is related to the proliferation and migration of hepatocellular carcinoma cells, and the recurrence rate of patients with high LKB1 expression is significantly increased after 5 years.
