摘要
为探明玉米及其野生近缘种大刍草对粘虫响应的异同,采用RNA-Seq技术对两者响应粘虫取食的转录谱进行探究。结果表明,玉米和大刍草被取食后上调的差异表达基因分别有1 755和1 761个,下调差异表达基因分别有3 309和1 723个。GO分析和KEGG分析表明,玉米和大刍草被取食后的上调差异表达基因较下调差异表达基因有更明显的差异,其中玉米中最显著上调的代谢通路与光合作用相关,而大刍草中则主要富集于植物激素信号转导、亚油酸代谢和MAPK信号通路。两者下调的差异表达基因均显著富集于核酸转录、翻译等过程。qRT-PCR验证结果与RNA-Seq测序结果一致。本研究从转录水平揭示了现代栽培玉米与其野生近缘种对害虫防御响应的不同。
To explore the different responses of maize(Zea mays ssp. mays) and teosinte(Zea mays ssp. parviglumis) attacked by armyworm, we adopted RNA-Seq technology to analyze their transcriptomes. The results showed that the number of up-regulated differentially expressed gene(DEGs) after attacked by armyworm were 1 755 and 1 761 in maize and teosinte, respectively, and the down-regulated DEGs were 3 309 and 1 723, respectively. GO function and KEGG pathway analysis showed that the up-regulated DEGs in maize and teosinte were more distinct than those of down-regulated DEGs. In the maize, the up-regulated DEGs were mostly associated with photosynthesis, while in the teosinte they enriched the process of plant hormone signal transduction, linoleic acid metabolism and MAPK signaling pathway. For the down-regulated DEGs, they all dominated the processes of transcription and translation in both maize and teosinte. The results of qRT-PCR agreed with the RNA-Seq from the aspect of transcriptome, for the first time, the present study revealed that the modern cultivated maize and its wild ancestor showed significant different responses by armyworm attack.
