摘要
以胭脂萝卜为试材,采用PCR扩增其ITS1-5.8SrRNA-ITS2序列,研究了序列同源性并构建系统发育树,以期为胭脂萝卜亲缘关系分析提供参考依据。结果表明:胭脂萝卜ITS1-5.8SrRNA-ITS2序列总长度为655bp,ITS1(G+C)含量为51.4%,长度为220bp;5.8SrRNA(G+C)含量为53.4%,长度为217bp;ITS2(G+C)含量为55.1%,长度为218bp。胭脂萝卜与台北的Mei-Hwa、南京的Cd-03、台北的Mei-Nong、南京的Yanghua、杭州的Baiyuchun、渥太华的SW77的同源性最高(99%),归为同一种。由系统发育树可得20个样品的总平均遗传距离是0.134,结合形态学特征表明待测胭脂萝卜可能由白皮白心萝卜进化而来。
To detect the genetic relationship of carmine radish,carmine radish was taken as the material,the complete ITS1-5.8SrRNA-ITS2 sequence was amplified by PCR,and homology analysis and phylogenetic tree were constructed.The results showed the length of ITS1-5.8SrRNA-ITS2 sequences was 655 bp,the(G+C)content of ITS1(220bp)was 51.4%,5.8SrRNA(217bp)was53.4%,ITS2(218bp)was 55.1%.The homology analysis showed carmine radish,Taipei(Mei-Hwa),Nanjing(Cd-03),Taipei(Mei-Nong),Nanjing(Yanghua),Hangzhou(Baiyuchun),and Ottawa(SW77)belonged to the same species with the highest homology(99%).The phylogenetic tree showed the average genetic distance of the 20 samples was 0.134.Combined with morphological features indicated that carmine radish may be evolved from radish with white episperm and white heart.
