摘要
真菌是药物先导结构的重要来源,得益于基因组测序技术的飞速发展,生物信息学分析发现真菌中存在大量次级代谢产物合成基因簇。这些基因簇在常规实验室培养条件下往往不表达代谢产物,被称之为"沉默"基因。尽管天然产物分离技术日益进步,结构新颖的天然产物发现几率却在减少。因此如何借助基因组数据分析并激活沉默基因簇,挖掘真菌潜在代谢潜能成为研究热点。本文主要介绍了基因组导向的丝状真菌沉默生物合成基因簇的激活策略。
Fungi are great sources of leading compounds.The significant advances of bioinformatics provide opportunities for obtaining more natural products than ever discovered.Genomic information has revealed a large number of fungal cryptic secondary metabolism gene clusters with unknown products.However,with the progress of natural product isolation and identification,the opportunity of finding new compounds has dramatically reduced.Thus,how to use the genomic data and activate the silent gene clusters to explore the metabolic potential of fungi has become a research focus.This paper gives an overview on the strategies for activating silent gene clusters in filamentous fungi by genomics methods.
