摘要
为了筛选与人类内源性逆转录病毒囊膜蛋白syncytin具有相互作用的蛋白质,首先分别构建诱饵质粒Syncytin-pKT25与基因组文库质粒,然后共转入报告菌株DHM1感受态细胞中,并采用抗性筛选得到含有互作蛋白质的菌株。结果发现,筛选到一条包括31个氨基酸的短肽。进一步通过回复杂交和Co-IP验证,确定筛选到的蛋白质与syncytin是否具有相互作用。实验结果表明,筛选到一条与syncytin具有相互作用的氨基酸短肽SJ-1。
In order to screen proteins that interact with the envelope protein syncytin of human endogenous retroviruses, the bait plasmid Syncytin-pKT25 and genomic library plasmids were constructed respectively, and they were co-transfected into the competent reporter strain DHM1 cells. After screening for antibiotic resistance, the strains containing the interacting proteins were identified and a short peptide consisting of 31 amino acids was found. The interaction between syncytin and the peptide was verified by recombination and Co-IP validation. The result demonstrated preliminarily that the short peptide named SJ-1 is an interacting peptide with syncytin.
