双酚S急性暴露对斑马鱼胚胎及子代胚胎发育的毒性效应 被引量:4

Acute toxicity effect of bisphenol S on zebrafish embryos and embryos of the next generation

摘要 目的研究作为双酚A替代物的双酚S(BPS)对斑马鱼的发育和生殖是否具有毒性效应。方法采用系列浓度的BPS对斑马鱼胚胎进行急性暴露,对胚胎活卵率,48 h和72 h孵化率,以及成鱼后处理组鱼与对照组鱼交配后子代胚胎的活卵率、48 h和72 h孵化率进行比较。结果 BPS处理斑马鱼胚胎,0. 001μmol/L、0. 01μmol/L、0. 1μmol/L和1μmol/L浓度对照组24 h活卵率平均数分别为81. 4%、78. 9%、79. 1%和83. 6%;而处理组24 h活卵率平均数分别为82. 2%、85. 2%、88. 1%和87. 3%。24 h死卵率对照组平均数分别为18. 5%、21. 0%、22. 8%和16. 3%;而处理组24 h死卵率分别为17. 7%、14. 7%、11. 9%和13. 3%。使用多因素方差分析,结果显示24 h活卵率和24 h死卵率差异有统计学意义(P <0. 05)。4个浓度对照组48 h孵化率平均数分别为0. 0%、7. 1%、29. 6%和69. 3%,而处理组平均数分别为48. 8%、40. 1%、28. 3%和38. 2%。统计分析结果显示48 h孵化率在组别和浓度双重影响下差异有统计学意义(P <0. 05)。BPS急性暴露后,胚胎发育成成鱼后再交配,不同浓度的BPS均对其子代胚胎的活卵率和孵化率产生了不同程度影响,其中48 h孵化率在每个浓度的各交配组间差异有统计学意义(P <0. 01)。纯对照组雌雄鱼交配后,4个浓度48 h孵化率分别为25. 0%、23. 2%、19. 8%和32. 4%;而纯处理组雌雄鱼交配后,48 h孵化率分别为76. 6%、49. 1%、32. 9%和22. 6%。结论斑马鱼胚胎在双酚S中急性暴露96 h,不仅斑马鱼胚胎发育受到影响,还首次发现其毒性效应具有可遗传性,即会影响其成鱼交配后子代胚胎的发育,尤其是孵化时间发生显著变化。 Objective To evaluate the toxic;ity effect of bisphenol S(BPS),substitute of bisphenol A on the embryonic,development and reproduction of zebrafish.Methods The zebrafish emhryos were exposed to different concentrations of BPS.The survival rate,hatching rate at 48h and 72h,the survival rate and the hatching rate at 48h and 72h of the next generation embryos produced by treatment group and the control group were compared.Results The survival rate of embryos were significant different between BPS-treatment groups and DMSO groups (P <0.05).The hatching rate of control group at 48h were respectively 81.4%,78.9%,79.1%and 83.6%at the concentrations of 0.001μmol/L,O.01μmol/L,0.1μmol/L and μmol/L; while that of the treatment group was respectively 82.2%,85.2%,88.1% and 87.3%;the death rate of embryos of the control group at 24h was respectively 18.5%,21.0%,22.8%and 16.3%;and that of the treatment group was respectively 17.7%,14.7%,11.9% and 13.3%.Multivariate analysis of variance showed that there was statistical significance on the difference of hatching rate and the survival rate at 24h hetween the two groups (P <O.05).At the four coneentrations,the hatching rates of the control group at 48h were 0.0%,7.1%,29.6%and 69.3%,respectively,while that of the treatment groups were 48.8%,40.1%,28.3% and 38.2%,respectively.Statistical analysis showed that the difference of hatching rate at 48h was statistieally significant between the two groups and among different concentrations (P <0.05).The embryos grew into adult fish and then mated.Different concentrations of BPS had different effects on the survival rate and hatc.hing rate of the offspring embryos.The differences of hatching rate at 48h had statistical significance at each concentration among different mating groups (P <0.01).After mating of male anct female fish in the control group,the hatching rates at four concentrations at 48h were 25.0%,23.2%,19.8%and 32.4%,respectively.And the hatching rates in the treatment group at 48h were 76.6%,49.1%,32.9%and 22.6%,respective
