摘要
目的 探究X射线联合低温等离子体(Non-thermal plasma, NTP)对人肺腺癌A549细胞以及人宫颈癌Hela细胞体外迁移及侵袭能力的影响及其可能的机制.方法 划痕实验以及Transwell小室法检测X射线及NTP对A549以及Hela细胞迁移及侵袭能力的影响;Western Blot法检测X射线及NTP对A549及Hela细胞Paxillin蛋白、FAK蛋白的磷酸化表达以及MMP-9蛋白表达的影响.结果 X射线联合NTP照射对A549以及Hela细胞的迁移(F=1 680.542、1 218.786,P<0.05)及侵袭(F=60.395、1 232.687,P<0.05)能力具有明显的抑制作用,且联合照射组强于X射线照射组(P<0.05),同时联合照射组对Paxillin以及FAK蛋白的磷酸化表达及MMP-9蛋白表达的抑制作用强于X线照射组.结论 NTP可提高X射线对A549及Hela细胞体外迁移及侵袭能力的抑制作用,其机制可能与下调Paxillin以及FAK蛋白的磷酸化以及MMP-9的表达有关.
Objective To evaluate the effects of X-ray combined with non-thermal plasma (NTP) on A549 and Hela cells. Methods Cells were divided into four groups, control group (Control), NTP treatment group (NTP), irradiation group (IR) and plasma plus irradiation group (NTP+IR). After treatment of plasma and irradiation, cell migration ability was detected by the scratch assay, cell invasion ability was detected by Transwell assay. Protein expression was detected by Western Blot assay. Results X-ray combined with NTP significantly suppressed the migration (F=1 680.542, 1 218.786, P<0.05) and invasion (F=60.395, 1 232.687, P<0.05) of A549 and Hela cells and its suppression effects were stronger than simple X-ray group (P<0.05). Moreover, the phosphorylation of Paxillin and FAK and the expression of MMP-9 were significantly suppressed by X-ray in combination with NTP, which were stronger than simple X-ray group. Conclusion NTP can enhance irradiation-induced suppression of migration and invasion of A549 cells and Hela cells. The suppression could be associated with the down-regulation of the expression of MMP-9 and the phosphorylation of Paxillin and FAK.
