利用γδ TCR四聚体检测分析外周血中CD14+CD277+单核-巨噬细胞的比例及其与治疗转归的关系 被引量:1

摘要 目的 探讨外周血中CD14+单核-巨噬细胞在γδ T细胞识别磷酸化抗原中的作用及其与治疗转归的关系.方法 应用3种γδ TCR四聚体对肺结核患者外周血和新生儿脐带血单个核细胞进行流式染色,分析各类抗原提呈细胞( APC)所占比例,根据临床病例分析其与治疗转归的关系.结果 在结核患者外周血和脐带血中同时与CD277抗体、γδ TCR四聚体结合比例最大且结合力最强的细胞是CD14+单核-巨噬细胞,其百分率中位数( P50)值在抗结核治疗1个月时达到最高峰值,综合临床病症结果显示该时段患者病情明显好转.结论 CD14+单核-巨噬细胞在γδ T细胞识别磷酸化抗原的各类提呈细胞群中所占比例最大,为深入探讨γδ T细胞限制性识别磷酸化抗原机制及同时参与固有免疫与适应性免疫提供了实验资料. Objective To investigate the role of peripheral CD14+monocyte-macrophages in the recognition of phosphorylated antigen by γδ T cells and its relationship with treatment outcome. Methods Three kinds of γδ TCR tetramers were used to stain PBMC collected from patients with tuberculosis ( TB) and neonatal umbilical cord blood samples. The proportions of various TB-specific antigen presenting cells (APC) in peripheral blood were analyzed, and their relationships with treatment outcome were assessed based upon clinical data. Results CD14+monocyte-macrophages both in tuberculosis patients′ peripheral blood and neonatal umbilical cord blood were the strongest binding cells to CD277 antibody and γδ TCR tet-ramers. The median (P50) of CD14+monocyte-macrophages reached the highest peak after taking anti-tu-berculosis treatment for about one month and patients′condition was improved obviously during this period. Conclusion This study elucidated that CD14+monocyte-macrophages accounted for the largest proportion of APC when γδ T cells recognized phosphorylated antigens, which provided reference data for further study on the mechanism of γδ T cells restrictively recognizing phosphorylated antigen and their significance in innate and adaptive immunity.
