摘要
从东北地区采集的150份猪肺脏样品中,检测出3株猪细小病毒毒株。根据PPV7标准株NCBI登录号KU563733设计3对引物,对3个病毒株的基因片段进行扩增测序,将测序结果依次进行拼接,获得3个病毒株的NS1和CP基因序列,然后进行序列的同源性比对及进化树分析。结果显示:3个毒株的NS1基因序列与经典PPV7毒株的同源性分别为94.4%~95.2%、94.1%~95.1%和93.5%~94.3%;CP基因序列与经典PPV7毒株的同源性分别为89.6%~91.8%、89.8%~92.0%和89.6%~91.7%。通过进化树比对分析,确认3株PPV毒株为PPV7亚型。通过对其他病毒进行检测,发现PPV7与PCV2、PPV2的混合感染率较高。该分析结果为我国东北地区猪病防控提供了依据。
Recently, Three strains of porcine parvovirus (PPV) were isolated from 150 pig lung samples collected from northeast China. Based on the reference sequence of PPV7 standard strain published in NCBI (accession number: KU563733), three pairs of primers were designed, and the gene fragments of the three virus strains were amplified and sequenced. After sequence assembly, the whole sequences of NS1 and CP gene of the three strains were obtained. At last, the sequence homology was compared, and evolutionary tree was drawn. The results showed that the sequence homologies of the NS1 gene between the 3 strains and the classical PPV7 strain were 94.4% to 95.2%, 94.1% to 95.1% and 93.5% to 94.3%, respectively. The homologies of the CP gene between the 3 strains and the classical PPV7 strain were 89.6% to 91.8%, 89.8% to 92.0% and 89.6% to 91.7%, respectively. According to the phylogenetic analysis, the 3 strains were all identified as PPV7 subtype. Through detecting other viruses, it was found that the mixed infection of PPV7, PCV2 and PPV2 was relatively. The analysis results would provide a basis for prevention and control of swine diseases in northeast China.
