红景天素上调大鼠嗅球成纤维细胞生长因子及其受体机制的探讨 被引量:2

摘要 目的:明确大鼠嗅球中成纤维细胞生长因子(FGF)合成、分泌和作用的靶细胞及其与受体(FGFR)表达的相关性;探讨中药红景天素抗大鼠嗅球衰老的信号传导机制。方法:取Wister大鼠青年对照组;老龄对照组和老龄红景天素用药组各10只,3个月后断头处死,取嗅球,以原位杂交和免疫组化方法分别检测FGFmRNA和FGF、FGFR蛋白在大鼠嗅球中的表达。结果:随着年龄的增长,大鼠嗅球中FGFmRNA、FGF、FGFR的表达明显减少;青年对照组和老龄对照组差异显著(P<0.01=;老龄红景天素组FGFmRNA、FGF、FGFR明显高于老龄对照组(P<0.05)。并且FGFR与FGF的表达呈明显的正相关。结论:大鼠嗅球组织的生长,发育与成纤维细胞生长因子及其受体信号传导密切相关;推测红景天素主要通过诱导大鼠嗅球FGF的表达,上调其受体FGFR的信号传导途径,发挥其对嗅球的抗衰老作用。 Objective:To investigate the relationship of the expression of fibroblast growth factor( FGF)and FGF target cells with FGF receptor(FGFR),the signal transduction mechan ism by which rhidosin inhibits senile effects was studied on the olfactory bulb in rats.Methods:Thirty rats were divided three groups:young control group(A),se n ile rhidosin group(B)(15mg rhidosin im x 90 days) and senile control group(C)( p hysiological saline).After removal of the heads of these rats,the olfactory bulb s were immediately fixed.The expressions of FGF,FGFR protein and FGFmRNA were de termined by immunohistochemical staining and situ hybridization.Results:The expr essions of FGFmRNA,FGF and FGFR on olfactory bulb gradually declined with ag ing.The positive expressions rate of FGFmRNA,FGF and FGFR were 76.1±5.20%,72 .5±±4.66% and 80.24±5.01% in group A;62.4±4.8%,58.6±3.42% and 60.8±3. 26% in group B;20.0±1.03%,18.2±0.94% and 24.36±1.50% in group C respectively ,wh ich showed significant difference between the group B and group A,group C(P<0 .01).Conclusion:FGF and its receptor signal transduction were closely related with the growth of olfactory cells.The Medicine Herbal rhidosin should be increase the expression of FGF on olfactory bulb in rats and up-regulate FGFR signal transduction.
