摘要
目的:研究制备抗人喉癌/抗蓖麻毒素A双功能抗体用以导向喉癌的治疗。方法:在制备抗人喉癌单克隆抗体杂交瘤细胞基础上,通过二次杂交瘤技术制备能够分泌抗人喉癌和抗蓖麻毒素A的杂交瘤细胞株。结果:该双功能抗体对Hep-2细胞株杀伤力比化学偶联法的双功能抗体强15倍。结论:采用二次杂交瘤技术制备的双功能抗体比化学偶联法的细胞毒性作用显著增高,优于化学偶联法。
Objective: To study the preparation of bifunctional antibody against human laryngeal cancer/anti-castor toxin-A and its cytotoxity activity for tumor cell-Methods:On the basis of preparation in monoclonal antibody against human laryngeal carcinoma,the hybridoma cell of anti-human laryngeal carcinoma/anti-castor toxin-A was prepared by the technique of secondary hybridoma.The secret bifunction antibody of the secondary hybridoma cell line was demonstrated by ELISA method.Result:The killing effect of the bifunctional antibody to Hep-2 cell line was 15 times stronger than that in the chemical coupling bifunctional antibody.Conclusion:Contrasting with chemical coupling bi functional antibody,the cytotoxity effect of bifunctional antibody which prepared by the technique of secondary hybridoma was obviously increased.The techniqueof secondary hybridoma was superior to one of chemical coupling bifunctional antibody.
