摘要
目的 :探讨人肿瘤坏死因子_α(hTNF_α)基因转染对舌癌细胞生长的影响。方法 :制备和纯化含hTNF_α基因的穿梭质粒 ,用DOSPER阳离子脂质体介导转染Tca8113舌癌细胞 ,对照组只给予等量的脂质体 ,不加入质粒 ;转染基因 2 4、4 8、72、92h后用ELISA法检测细胞培养上清液中hTNF_α的浓度 ,并用MTT法检测细胞的存活率。结果 :基因转染 2 4、4 8、72、96h后转染组与对照组细胞培养上清液中hTNF_α浓度之间的差异均有显著性 (P <0 0 5 ) ,转染组各个时期浓度均高于对照组 ;MTT法检测相应各时期转染组与对照组平均OD值之间的差异均有显著性 (P <0 0 5 ) ,转染组各个时期平均OD值均低于对照组 ,转染细胞的存活率降低 ,生长受抑制。结论
Objective: The aim of this study was to investigate suppression effects of the transfection of human tumor necrosis factor_α (hTNF_α) gene on tongue carcinoma cells.Methods: The shuttle plasmid containing hTNF_α gene was extracted and purified, then it was transferred into Tca8113 tongue carcinoma cells with cationic liposome DOSPER. The control group was only given equivalent liposomes, except the plasmid. After culturing for 24, 48, 72 and 96 hours, the expression of hTNF_α gene in Tca8113 cells was analyzed by ELISA and, the survival rate of transferred cells was assayed by MTT enzymatic labeling technique.Results: The transferred Tca8113 cells displayed significantly overexpression of hTNF_α (P<0.05). The survival rate of the transferred Tca8113 cells was decreased significantly (P<0.05).Conclusion: Transfection of hTNF_α gene in vitro mediated by cationic liposomes can induce the overexpression of hTNF_α and inhibit the growth of tongue carcinoma cells.
