摘要
目的 :研究端粒酶的 3种组分与其活性的关系 ,探讨端粒酶激活的关键因素。方法 :采用 TRAP法检测大肠癌组织及相邻正常黏膜组织中端粒酶活性 ,用 RT-PCR检测端粒酶 3种组分的表达。结果 :在 6 4例 ( 85 .33% )癌组织中检测到端粒酶活性 ,正常黏膜中没有端粒酶活性 ,两者差异有显著性 ( P <0 .0 1) ;h TR和 TP1基因的表达在癌组织和正常黏膜没有差别 ,而 h TERT在癌组织中的表达要明显高于正常黏膜 ,差别具有显著性 ( P <0 .0 1) ,h TERT基因表达强度与端粒酶活性密切相关。结论 :大肠癌端粒酶活性表达具有肿瘤特异性 ,h TERT在癌组织中的表达明显高于正常粘膜 ,大肠癌中端粒酶活性和 h TERT基因表达强度密切相关 ,h
Objective: To investigate the relation between the three components and the activity of telomerase and to find out which component is the rate limiting determinant of the enzymatic activity Methods: Using the telomerase repeat accelerate protocol (TRAP) to detect the telomerase activity and using the RT PCR to detect the expression of the three components of telomerase Results: More than 85% colorectal cancer tissues have telomerase activity whereas normal colon mucosa haven't hTR and TP1 mPNA were commonly expressed in cancer tissues and normal mucosa However the expression of hTERT in Cancer tissues are much higher than in the normal tissues The difference is significant There is strong correlation between telomerase activity and hTERT expression Conclusion: Telomerase has a special expression in colorectal cancer The expression of hTERT is higher in cancer tissues than in the normal tissues The expression of hTERT may be the rate limiting determinant of the enzymatic activity
