摘要 目的 :应用光镜及免疫组织化学技术观察转基因糖尿病小鼠颌下腺的形态学改变以及血管内皮生长因子 (Vascularendothelialcellgrowthfactor ,VEGF)在颌下腺表达的变化 ,旨在说明糖尿病时颌下腺功能损害与上述因子间的关系。方法 :引进日本C5 7BL/ksj—db/ +m表型正常隐性基因小鼠 ,近亲交配 ,其纯合子后代 ,即为db/db(单基因遗传自然发病型 )糖尿病小鼠。取 3、4、6、8、10月龄db/db糖尿病小鼠及相应年龄段的db/ +m正常小鼠 (对照组 )颌下腺 ,常规固定包埋 ,切片 ,行HE及SP免疫组化染色 ,图象分析仪计数颌下腺内VEGF免疫阳性的细胞数。结果 :随着糖尿病进展 ,颌下腺组织萎缩 ,但间质增生 ,可见血管增多。在db/db糖尿病小鼠VEGF阳性细胞数随着糖尿病进展有一明显增多趋势 ,且与各同龄对照组相比明显增强 (p <0 .0 5 )。结论 :VEGF在糖尿病颌下腺表达增强,可能与缺氧有关 ,进而导致间质血管增生 ,新血管形成 ,这也是糖尿病性微血管病变的相关因素之一。 Objective: To investigate the morphological changes and expressions of relative cell factor, vascular endothelial cell growth factor (VEGF), in C57BL /ksj -db/db mutant diabetic mice submandicular gland. At the same time, the present study is aimed at clarifying the factor's roles in diabetic animal submandicular gland dysfunction, then can supply experimented data for the found and clinical study on diabetes millitus.Method: Light microscope and immunochemistry technologies were adopted in the study.Animals were C57 bl/ksj-db/+m mutant diabetic mouse which were imported from Japan. The pure zygote, that is db/db mouse, displays obesity and high blood glucose beyond 10mmol/l. The test group selected 3,5,6,8,10 mouth-old db/db mouse. The age -matched db/+m mouse as controls. The experimental data were statistical analysis.Results: In db/db diabetic mice, submandicular gland atrophied and took on the significant morphological changes in acinar,duct and granular convolted tubule. Immunohistochemistry results as follow:① VEGF was expressed in submandicular gland from both tests and control mice.②Immunopositive cell of VEGF was significant increased in diabetes mice than those of control, and there was an enhance decline as disease progressed in DM groups. Conclusion: VEGF immunopositive cell was increased with development of disease, which could associate with the tissue oxygen deficience, therefore, result in microvascular proliferation and new vascular formation.
