摘要
本文为以Fmoc(Nα-芴甲氧羰基 )固相合成法制备的胸腺素α1粗品提出了一条实验室规模的液相色谱法纯化路线 ,共分两步 :第一步采用DEAESepharoseFastFlow填料进行离子交换层析操作 ,分离除去粗品中的大部分杂质 ;第二步用制备型反相高效液相色谱 (用C1 8色谱柱 )进行最后的纯化。终产物纯度可达 95% ,生物活性符合实验指标。
In this paper,a purification route using the liquid chromatogram on a loboratory scale was established on thymosin α1 synthesized by the solid-phase synthetic method using Fmoc group fo protecting α-amino acid.Two steps were included in the purification route.First was the Ion-exchange chromatography (IEC) using DEAE Sepharose Fast Flow as the matrix.After this step,majority of impure peptides was removed.Second was the preparation RP-HPLC on C18 column.Then the final product's purity attained to 95% and the values of the biological activity all meet the requirements.Those operation conditions during the separation and purification process were also studied and optimized in this thesis.
