摘要
为研究探讨吉林西部草原鼠疫菌生物学性状,本研究对该地区30株鼠疫菌的生化性状、质粒种类及质粒DNA酶切图谱、染色体PCR指纹图谱和外膜蛋白种类进行了测试。通过系统研究,证实该地区鼠疫菌生化结果为,分解葡萄糖、麦芽糖和甘油,不分解乳糖、蔗糖、山梨醇、鼠李糖、密二糖和尿素,硝酸盐还原(脱氧)试验阳性,靛基质试验阳住,V-P试验阴性,M-R试验阳性;试验全部菌株都具有规范的质粒图谱,携带的质粒种类为,65Mdal、45Mdal、6Mdal3种;被试菌株的质粒单纯用EcoRI酶切,可切成16个片段;鼠疫菌染色体PCR指纹图谱分析,吉林西部草原场区鼠疫菌的染色体可产生8条主要DNA带;可产生15条外膜蛋白带。
In order to study the biological character of Y. Pestis in the west part in Jilin Province, 30 strains of Y. Pestis in this area were tested for their biochemical character, plasmid class, endonuclease digest map of plasmid DNA, PCR finger print map of chromosome and the class of out membrane protein. Through the systemic study, the biochemical results of these strains in this ares were following. These strains of Y. Pestis utilized glucose, maltose and glycerin, but they didn't utilize lactose, sacckarose, sorbit, rhamnose, melibiose and urea; Nitrate reducing, indole, M - R test were postive, V - P test was negative. All the strains tested have standud plasmid map, they carry three kinds of plasmid, 6 Mdal ,45 Mdal, and 65 Mdal; These plasmids were cut by singl Enzyme EcoR I into 16 segments. S main bands were found in the chromosome of Y. Pestis in the Base area of the west prairie in Jilin Province by the analysis of PCR finger print of the chromosome of Y. Pestis. 15 out membrane protein bands were created.
