中枢白细胞介素-1受体参与应激镇痛

摘要 目的 :观察中枢白细胞介素 1(interleukin 1,IL 1)受体在足击镇痛及条件恐惧应激刺激诱发镇痛中的作用。方法 :本实验用脑室注射 (intracerebroventricularinjection ,icv)生理盐水 (normalsaline ,NS)或白细胞介素 1受体拮抗剂 (interleukin 1receptorantagonist,IL 1ra)并用热辐射甩尾测痛法检查IL 1ra对足击镇痛及条件恐惧应激刺激诱发镇痛效应的影响 ;结果 :(1)icvNS的大鼠 ,足击引起痛阈上升 ;而icvIL 1ra 30min后足击不引起明显的痛阈上升 ,但icvIL 1ra 90min后足击仍出现明显的痛阈上升。(2 )icvNS的大鼠 ,条件恐惧应激刺激也引起明显的痛阈上升 ,且该条件恐惧应激刺激诱发的镇痛效应能被icvIL 1ra明显衰减。 (3)icvIL 1ra本身对基础痛阈无明显影响。结论 :中枢IL 1受体参与足击及条件恐惧应激刺激诱发的镇痛效应。 Objective: To test the effects of introcerebroventricular injection of (icv) interleukin 1 receptor antagonist (IL 1ra) on analgesia induced by footshocks or conditioned fear stimuli. Methods: The influence of icv IL 1ra on analgesia induced by footshocks or conditioned fear stimuli was observed by measuring the tail flick latency in SD rats. Results: although footshocks induced an increase in pain threshold in normal saline (NS) injected rats, no significant increase in pain threshold could be induced if footshocks were given to the rats 30 min after icv IL 1ra. Interestingly, if the rats received footshocks 90 min after icv IL 1ra, footshocks significantly increased the pain threshold. In addition, analgesic effects were also induced by conditioned fear stimuli,which could be attenuated by icv IL 1ra. In our experiments, icv IL 1ra had no significant influences on pain threshold. Conclusion: Above results suggest that central IL 1 receptors are involved in analgesia induced by footshocks or conditioned fear stimuli.
