摘要
目的研究脑胶质瘤中Th1/Th2类细胞因子的不均衡表达及其意义。方法以IFN-γ、IL-2和IL-4、IL-6、IL-10、IL-13分别代表Th1和Th2类细胞因子,采用半定量逆转录聚合酶链反应技术检测上述细胞因子基因在62例脑胶质瘤组织、4株脑胶质瘤细胞株、15例脑胶质瘤患者外周血单个核细胞以及人正常脑组织、良性脑膜瘤组织中的表达情况。结果脑胶质瘤组织和脑胶质瘤细胞株Th2类细胞因子的表达明显占优势(P<0.01),胶质瘤患者外周血亦表现出Th2类细胞因子优势表达的趋势,而人正常脑组织和良性脑膜瘤组织中两类细胞因子的表达差异无显著性。结论脑胶质瘤中Th2类细胞因子的优势表达可能与肿瘤的发生发展及免疫逃逸有关。
Objective To study the significance of the unbalanced expression of Th1/Th2type cytokines in human glioma.Methods The gene expressions of Th1/Th2type cytokines in62specimens of human glioma tissues,4glioma cell lines,peripheral blood mononuclear cell(PBMC)of15glioma patients,5specimens of normal adult brain tissue and5brain meningioma tissues were detected by semiquanti-tative reverse transcription polymerase chain reaction.IFN-γand IL-2represent Th1type cytokines.IL-4,IL-6,IL-10and IL-13represent Th2type cytokines.Results There were obviously predominant expre-ssion of Th2type cytokines in glioma cell lines(P<0.01)and specimens of human glioma tissues(P<0.01).The tendency of distinct expression of Th2type cytokines in PBMC was also existent.There wasn' t obvious discrepancy of the expression of two type cytokines in normal adult brain tissues and meningioma tissues.Conclusions It is likely that the switching of Th1/Th2type cytokines in gliomas as predominant expression of Th2type cytokine genes is related to the origination of gliomas and the evasion of glioma cells from immune surveillance.
