摘要
目的 优化关节软骨组织工程种子细胞的体外获取方法。方法 分析不同代次体外单层培养幼兔关节软骨细胞接种密度与相对接种存活率的关系 ,并观察其细胞生长状况、形态特征及分化功能。结果 相同培养条件下 ,体外单层培养关节软骨细胞的相对接种存活率和生长活性与其接种密度呈正相关 ;同一代次适宜密度 [( 2~ 3)× 10 4 cm2 ]接种者的相对接种存活率和生长活性同高密度 [( 4~ 5 )× 10 4 cm2 ]接种者相近 ;传代培养 (第 2~ 4代 )细胞的相对接种存活率和生长活性相对高于原代培养者。结论 在相同培养条件下及一定传代次数内 ,适宜密度接种进行关节软骨细胞单层传代培养 ,可获取数量多。
Objective To optimize the accumulation method of seeding cells for articular cartilage tissue engineering. Methods Relation between the plating density and the plating efficiency of young rabbit articular cartilage cells at different passages in monolayer culture were analyzed, and the growth status, morphologic features, and differentiation properties of those cells were observed. Results Under the same circumstances, the plating efficiency, the growth activity of primary and passage articular cartilage cells in monolayer culture significantly related to the plating density (The larger the density was, the higher plating efficiency and activity of the cells were). The plating efficiency and growth activity of the cells at the appropriate plating density (2~3×10 4 cells/cm 2 ) was close to those in the high plating density, and the plating efficiency and grouth activity of cell in passage culture (passages Ⅱ~Ⅳ) were higher than those of primary cells.Conclusions After the division and proliferation of articular cartilage cells in monolayer culture at the appropiate plating density, a large number of seeding cells with high growth activity can be obtained for articular cartilage tissue engineering.
