摘要
目的 研究槲皮素对体外培养的SD大鼠破骨细胞增殖及凋亡的影响。方法 体外机械分离SD大鼠破骨细胞 ,在空白对照及含槲皮素 2 .5、10、4 0mmol·L-1的 4种MEM培养基中进行培养 ,并于 4 8、72、96、12 0h涂片经瑞氏染色和TUNEL法染色观察各培养孔内破骨细胞数目、形态、凋亡情况。结果 各组破骨细胞数目随着药物浓度增加而减少 ,P <0 .0 0 5。药物浓度的增加使破骨细胞凋亡的比例呈增加趋势。结论 槲皮素能够明显抑制大鼠体外培养的破骨细胞增殖 ,并存在诱导其凋亡的作用。
OBJECTIVE To study the effects of quercetin on proliferation and apoptosis of SD rat osteoclasts in vitro .METHODS Osteoclasts and their precursors of SD rats were mechanically isolated from long bones.The cells were cultured in four classes of minimum essential medium(MEM),which quercetin concentration was 0, 2.5, 10,40 μmol·L -1 , respectively. Dynamic changes in morphology, numbers and apoptosis of the osteoclast were detected with Wright's or TUNEL staining at 72, 96, 120 and 144 h from beginning of culture.RESULTS The number of osteoclast was decreased,but the number of apoptic osteoclast had a tendency of augmentation with increasing concentration of quercetin.CONCLUSION Quercetin could inhibit the proliferation and induce apoptosis of osteoclasts cultured in vitro .
