摘要
目的 :研究磁导向下磁性药物载体脑定向分布 ,磁性药物载体对肝、肾及骨髓的毒副作用 ,为脑胶质瘤的磁导向化疗提供可靠的依据。方法 :将合成的磁微球 白蛋白 氨甲喋呤载体 (FM HAS MTX) ,经尾静脉注入 30只SD大鼠体循环内 ,在大脑右侧放置一个梯度磁场 ,观察不同时间 ( 15、30、45min)时氨甲喋呤 (MTX)在SD大鼠大脑的分布状况以及相同时间内左右大脑内MTX含量比较 ,同时研究磁导向与非磁导向时脑内MTX含量变化。另取昆明小鼠 5 0只 ,将一定的磁性药物载体混悬液通过尾静脉注入小鼠体内 ,头部一侧放置聚焦磁场 ,定期给药观察 3个月 ,定时取血检查红细胞、白细胞、血小板、谷丙转氨酶、非蛋白氮 ,并设立对照组。结果 :45min时磁导向组右侧大脑MTX含量 ( 0 5 6± 0 0 2 )是非磁导组右侧大脑MTX含量 ( 0 0 6± 0 0 2 )的 9 3倍 ,不同时间内右大脑 (磁导向侧 )与左侧大脑MTX含量比较有显著性差异 (P <0 0 1) ,相同时间内 ,右侧大脑MTX含量明显高于左侧大脑(P <0 0 1)。 7、14d磁导向组红细胞为 ( 935 0± 79 2 )、( 92 5 0± 81 5 )万 /mm3,肝功能及肾功能变化不大 ,非磁导向组 (MTX组 )红细胞为 ( 5 70 0± 82 2 )、( 5 0 5 0± 81 2 )万 /mm3,肝功能及肾功能变化较大 。
Objective: To study the targeted brain distribution of FM HSA MTX carriers under magnetic guidance and side effect of the carriers on liver,kidney and bone marrow. To provide reliable evidence for magnetic targeting chemotherapy of malignant glioma. Methods: FM HSA MTX were injected into systemic circulation of 30 SD rats through caudal vein. A magnetic field was located at the right brain. MTX distribution in brain was observed at the same points. Meanwhile, the changes in MTX contents in brain was studied in the presence or absence of the magnetic field. Fifty Kunming mice were used. FM HSA MTX carriers were injected into the mice regularly via tail vein. Magnetic field was placed near the right brain of mice. The mice were observed for 3 months. The mice blood was taken regularly to determine RBC, WBC, BPC,ALT and NPN were checked respectively. Results: MTX contents of the right brain were studied in the presence of magnetic field 0.56±0.02 was 9.3 times as many as that of the right brain in the absence of magnetic field 0.06±0.02 at 45 min. There was significant difference in MTX contents between the right and left brain at different time points ( P <0 01). At the same time point, MTX contents of the right brain was significantly higher than that of the left brain ( P <0.01).At 7 d and 14 d,RBC (935.0±79.2),(925.0±81.5),ALT, NPN had no obvious change in magnetic targeting group (FM HSA MTX group),Blood cell and ALT with MTX group,while RBC (570.0±82.2),(505.0±81.2),ALT and NPN changed evidently in MTX group ( P < 0.01). Conclusion: Under magnet guidence, FM HSA MTX carriers was distributed to brain targeting area, which may change the characteristics of MTX flow distribution.The collection of MTX was reduced in liver, kidney and bone marrow,alleviating the damage to them.
