摘要
目的探讨在高三尖杉酯碱 (HHT)诱导K5 6 2细胞凋亡时 ,bcl- 2家族部分基因在mRNA水平表达的改变。 方法细胞形态 (光镜和电镜 )、DNA凝胶电泳、流式细胞仪等方法证实凋亡 ,进一步用RT -PCR方法研究凋亡过程中bcl- 2家族中部分基因表达的改变。 结果超过一定“阈浓度”HHT可诱导K5 6 2细胞凋亡 ;RT -PCR方法发现bax基因和bfl- 1基因在加药 6h开始上调 ,家族其它基因bcl- 2、bcl-xL的表达未见明显改变。 结论HHT可诱导K5 6 2细胞凋亡 ;其机制可能与HHT在mRNA水平上调bax基因有关 ;而bfl- 1基因的上调可能是K5 6
Objective To study the effects of bcl-2 family in apoptosis induced by homoharringtonine on K562 cells. Methods Cell morphology, DNA gel electrophoresis, flow cytometry and RT_PCR were used in this study. Results Apoptosis was detected on K562 cells when the concentration of HHT reached a certain threshold. RT_PCR showed that the bax gene and bfl_l gene were up_re_ gulated gradually at 6h after HHT treatment, but the other two genes in the family: bcl_2 and bcl_xL, had no change through this procedure. Conclusion HHT may induce K562 cell apoptosis. The up_regulation of bax gene perhaps contributes to the procedure induced by HHT. bfl_1 the new member of bcl_2 family, its gradually up_regulation probably may be the reason for K562 cell to resist further apoptosis induced by HHT.
