视黄酸受体在脐血淋巴细胞细胞因子表达中的调节作用 被引量:2

摘要 目的 研究视黄酸 (RA)促进脐血淋巴细胞 (CBL)细胞因子表达的作用途径 ,探讨视黄酸受体α在该过程中的可能作用。方法 收集 19例健康新生儿的脐血标本进行体外培养 ,采用RT PCR法检测CBL中视黄酸受体α(RAR α)、IL 2、IFN γ和IL 4的mRNA水平。结果 RA上调CBL中RAR α的基因表达水平 ,培养 2 4小时时的上调幅度为 (1 17± 0 0 7)倍 (P <0 0 0 1)。RA上调IL 2的基因表达水平 ,培养 2 4小时时的上调幅度为 (1 2 9± 0 2 0 )倍 (P <0 0 0 1)。RA也上调IFN γ的基因表达水平 ,培养 4 8小时时的上调幅度为 (1 39± 0 2 6 )倍 (P <0 0 0 1)。RA还上调IL 4的基因表达水平 ,培养 2 4小时时的上调幅度为 (1 5 2± 0 4 4 )倍 (P <0 0 0 2 )。RA使RAR α基因表达的上调幅度与RA使IL 2、IFN γ和IL 4基因表达的上调幅度皆存在正相关 ,r值分别为 0 4 0、0 39和 0 4 6 ,(P <0 0 5 )。结论 RAR α可能参与RA对CBL细胞因子 (IL 2、IFN γ和IL 4 )表达的促进作用。RA可能通过增加RAR α的表达而上调IL 2、IFN γ及IL 4等细胞因子的表达 。 Objective To explore the path of retinoic acid (RA) promoting cytokine expressions in cord blood lymphocytes (CBL) Methods Nineteen cord blood samples from healthy newborns were collected and CBL were cultured in vitro The mRNA levels of retonoic acid receptor α(RAR α), IL 2, IFN γ and IL 4 were determined by reverse transcription polymerase chain reaction(RT PCR) Results The RAR α expression was increased with the optimal concentration of RA, the enhanced amplitude was (1 17±0 07) times after culture for 24 hours ( P <0 001) The expression of IL 2 was augmented by RA, the enhanced amplitude was (1 29±0 20)times after culture for 24 hours ( P <0 001) The expression of IFN γ was augmented by RA, the enhanced amplitude was (1 39±0 26)times after culture for 48 hours ( P <0 001) The expression of IL 4 was augmented by RA, the enhanced amplitude was (1 52±0 44) times after culture for 24 hours ( P <0 002) There were positive correlations between the enhanced amplitude of RAR α expression and the enhanced amplitudes of IL 2?IFN γ and IL 4 expressions, r =0 40, 0 39, 0 46, respectively ( P <0 05) Conclusion RAR α might be involved in the promotive cytokine expressions in CBL by RA
