摘要
目的 观察重组人白介素10(rhIL-10)对血管平滑肌细胞(VSMC)增殖的影响。方法 离体培养大鼠胸主动脉 VSMC并分别接受不同浓度rhIL-10、肿瘤坏死因子(TNF-α)、血小板源性生长因子(PDGF-BB)的刺激,采用MTS/PES 法确定VSMC的增殖状态并与对照组比较。结果 与对照组相比,TNF-α和PDGF-BB均对VSMC增殖具有明显的刺 激作用(P<0.05)。单独应用rhIL-10对血管平滑肌细胞生长没有影响。在TNF-α和PDGF-BB分别刺激下,rhIL-10均可 抑制VSMC的生长(P<0.05)。结论 rhIL-10可抑制细胞因子和生长因子诱导的VSMC增殖。
Objective To observe the effects of recombinant human interleukin-10 (rhIL-10) on the proliferation of rat vascular smooth muscle cells (VSMCs) cultured in vitro. Method Aortic VSMCs cultured in vitro were treated with varied doses of rhlI-10 alone or in combination with tumor necrosis factor-a (TGF-a) or platelet-derived growth factor-BB(PDGF-BB) respectively. The VSMC proliferation was quantified by colormetric assay. Results Compared with control, both TNF-a and PDGF-BB stimulated conspicuous proliferation of VSMCS, bit the effect of which was not observed by rhIL-10 treatment alone. In the presence of rhlL-10, the proliferation effects of both TGF-a and PDGF-BB on VSMCs were significantly inhibited (P<0.05). Conclusion rhIL-10 can inhibit VSMC proliferation induced by TNF-a and PDGF-BB, thus may provide a new therapeutic approach for regulating vascular wall remodeling after vascular injury.
