摘要
采用10.000xg离心,55％硫酸胺沉淀和DEAE-SephadexA-50柱层析纯化了豚鼠腹腔单核细胞和吞噬细胞脂加氧酶,并研究了该酶的分子特性。该酶等电点4.5,分子量约76.7KDa。但采用SDS一梯度聚丙烯酰胺凝胶电泳时出现另一分子量约21.8KDa的蛋白质区带,提示此种酶可能由四条多肽链组成。
Lipoxygenase from guinea pig peritoneal monocytes/phagocytes was purified by several steps including 10,000×g centrifugation, ammonium sulfate precipitation and chromatography on DEAE-Sephadex A-50 column. The molecular characteristics were also studied. The results show that the isoelectric point of this enzyme is at about 4.5 and molecular weight is about 76.7 KDa. The enzyme maybe consists of four polypeptide chains because of the fact that another protein zone is found with molecular weight at about 21.8 KDa.
