摘要
采用离子交换层析 ,一步分离纯化鱿鱼肝脏中的核酸初抽提样品 ,层析条件 :填料为DEAE 纤维素 ,色谱柱 8cm× 1cm ,流速 1ml/min ,2 60nm紫外检测 ,收集以 0 6mol/LNaCl( pH 7 4Tris Cl缓冲液 )为洗脱相时的洗脱液 ,得率为 85 %~89 % ,经透析、低压冷冻干燥 ,可获得纯度为 87%~ 90 %的核酸产品 ,可用于医药及食品工业。
The ion exchange chromatography was used to purify nucleic acid from the primary nucleic acid sample of squid livers. The eluant was inspected by 260 nm UV monitor.Nucleic acid was eluted with 0 6 mol/L NaCl(Tris Cl pH 7 4)and selected.After dialysis and lyophilizing, 87%~90% pure product can be attained.The product can be used for medical industry and food additive.
