摘要
目的 :探讨烫伤后机体及创面区域巨噬细胞集落刺激因子 (M- CSF)水平的变化。方法 :采用 2 0 % 度烫伤模型 ,将48只小鼠随机分为烫伤组、假烫组 ,每组内根据取材时间不同分为 4个亚组。于烫伤后第 1、3、5、10天 ,分别取静脉血、腹部正常皮肤、肌肉、脾、肾、肝、肺、脑、创面区域组织或假烫组小鼠相应背部皮肤 ,用 EL ISA法检测 M- CSF水平。 结果 :M- CSF的表达在烫伤组、假烫组小鼠脑组织中均未测出 ,而其余组织中均可测出 ,但两组动物各组织 M- CSF水平无显著差别 ;小鼠烫伤创面区域的 M- CSF水平在伤后逐步升高 ,于伤后第 5、10天明显高于假烫组 [分别为 (2 .2 6± 0 .2 1) vs (0 .92± 0 .10 )及(4.96± 0 .36 ) vs (0 .88± 0 .13) pg/ m g组织 ,P<0 .0 5 ]。结论 :2 0 % 度烫伤后机体各组织 M- CSF水平的变化主要局限在创面局部。创面区域 M- CSF水平升高 ,可能与创面局部的成纤维细胞、内皮细胞有关。
Objective: To study the changes of macrophage colony-stimulating factor (M-CSF) in organs and wounds in the mice with 20% TBSA full-thickness burn injury. Methods: Forty-eight pathogen-free C57BL/6 mice were randomly divided into BURN and SHAM group. In BURN group, each mouse had a 20% full-thickness burn injury. The blood, abdominal skin, muscle, spleen, kidney, liver, lung, brain and burned skin were removed and cultured at day 1, 3, 5 and 10 after injury respectively. Control samples were obtained from SHAM in the comparable sites. The concentration of M-CSF in the cultured media were measured by ELISA. Results: The M-CSF levels of burned skin increased gradually after burn injury. At day 5 and 10, the M-CSF levels of burned skin were (2.26±0.21) pg/mg and (4.96±0.36) pg/mg respectively. Conclusion: TBSA full-thickness burn injury (20%) up-regulates the protein levels of M-CSF in burned skin, suggesting that M-CSF may contribute to wound healing in injured skin.
