摘要
目的 研究抗纤灵的抑癌作用及分子机制。方法 运用血清药理学及免疫组织化学等方法 ,观察抗纤灵药物血清对人肝癌细胞 (Hep G2 )的体外生长抑制作用及对 P2 1 WAF 1 /CIP1蛋白表达的影响。结果 含抗纤灵血清 18,9,3g/ kg对体外培养的 Hep G2 细胞的细胞生长抑制率 (% )分别为 6 3.0 9± 5 .0 8,5 5 .0 9± 4.0 9,48.0 8± 4.0 8,细胞毒活性 (% )分别为 48.0 6± 4.0 8,38.0 9± 5 .0 1,2 5 .0 0± 4.13,其细胞毒活性均高于空白血清 (P<0 .0 1)。含抗纤灵血清各组细胞生长抑制率和细胞毒活性与环磷酰胺血清组比较 ,差异有显著性或无显著性。含抗纤灵血清 18,9,3g/ kg各组 Hep G2 P2 1 WAF1 /CIP1 蛋白表达高于环磷酰胺血清组及空白血清组 (P<0 .0 1)。结论 抗纤灵具有抑制人肝癌细胞体外生长的作用 ,其分子机制与该方提高 Hep G2 P2 1 WAF 1 /CIP1
Object To study the antitumor action of KANGXIANLING (KXL), a traditional herbal preparation and its mechanism of action at molecular pharmacological level. Methods Effect of KXL containing serum on cell periferation and protein expression of P 21 WAF1/CIP1 of human hepatic cancer HepG 2 were studied by serologic pharmacologic tests. Results KXL containing serum at concentrations of 18, 9, 3 g/kg inhibited proliferation of ex vivo cultured HepG 2 cells by (63.09±5.08)%,(55.09±4.09)%, and (48.08±4.08)% respectively. Their cytotoxicities were (48.06±4.08)%, (38.09±5.01)%, and ( 25.00± 4.13)% respectively, which were all higher than that of blank serum without KXL (P<0 01). When compared with cyclophosphamide (CTX) containing serum, their inhibition of cell proliferation and cytotoxicities showed either statistically significant or non significant differences. Protein expresion of HepG 2 P 21 WAF1/CIP1 were higher than that of CTX containing serum or blank serum (P<0 01). Conclusion KXL could inhibit proliferation of HepG 2 cells cultured ex vivo relevant to its increased protein expression of HepG 2, P 21 WAF1/CIP1 .
