摘要
应用圆二色谱 (CD谱 ) ,测定了小麦多聚半乳糖醛酸酶抑制蛋白 (PGIP)在不同pH和温度影响下的构象变化 ,并和生物活性作了比较 .天然小麦PGIP的CD谱显示 2 11nm处的单一负峰 ,此时小麦PGIP含有较多 β 折叠(4 3.7% )和 13.1%α 螺旋 ,这是关于小麦PGIP二级结构的首次报道 .pH和温度引起小麦PGIP的活力改变与CD谱的变化有密切的关系 .活性降低时 ,CD谱峰峰值变小 ,活性完全丧失时 ,负峰蓝移 .根据给出的 [θ]值计算结果 ,发现 β 折叠与小麦PGIP的活性直接相关 .图 2表 2参 2
The activity and conformation changes of polygalacturonase inhibiting protein (PGIP) from wheat denatured by pH and temperature was studied by the measurement of circular dichroic (CD) spectra. The native PGIP of wheat exhibited a characteristic CD spectral negative band centered at 211 nm. It was estimated to contain about 43.7% β sheet and 13.1% α helix. This is the first study on the secondary structure of wheat PGIP. With the change in activity of wheat PGIP induced by pH and temperature, CD spectra changed. The band was smaller when activity was weaker, and it had blue shift while wheat PGIP was inactive. The further studies showed that β sheet was directly interrelated with the changes of activity of PGIP from wheat.Fig 2, Tab 2, Ref 23
