摘要
应用原位杂交技术 ,研究了大鼠出生后发育过程中 ,听皮层GABAA受体α2及β3亚单位mRNA年龄相关的表达变化。特异性DIG标记寡核苷酸探针检测显示 ,α2亚单位mRNA阳性神经元数量从出生后第 1天开始逐渐增加 ,到第 12d达到一个高峰 ,而后逐渐减少 ,出生后 2 1d降至检测水平以下 ;β3亚单位mRNA阳性神经元数量从出生后到第 12d ,一直保持较高表达水平 ,第 12d后急剧降低 ,第 2 1d后降至检测水平以下。研究结果为进一步在皮层水平上探讨出生后听觉功能发育可塑性的分子机制提供了重要资料。
Using in situ hybridization technique, we studied the age related mRNA expression changes of GABA A receptor α2 and β3 subunits during postnatal development in the auditory cortex of the rats.By specific DIG labeled oligo nucleotide probes detected the results showed that the number of α2 positive cells significantly increased from postnatal day (PND)1 rat pups, peaked on PND 12 and then declined markedly.On PND 21 the expression level was very low.β3 subunit expression kept a high expression from PND 1 to PND 12 and then dropped rapidly. The β3 subunit expression could not be detected on PND 21.The results provide important datas for investigating the molecular mechanism of development plasticity of auditory function at cortex levels. \[
