摘要
制备食管癌细胞核基质,以其为抗原建立了Elisa检测法。结果表明,这种核基质不仅具有较好的抗原免疫性,还具有良好的抗原反应性。在我们制备的小鼠体内克隆细胞株的腹水中,以5ug抗原蛋白量包被,抗体滴度达1/2000也能获得满意的结果。本法不仅为筛选食管癌单克隆抗体提供了一种灵敏、方便的手段,也为今后临床应用打下基础。
An enzyme-linked immrnosorbent assary(ELIAS)was developed for identification and puantitation of nuclear matrix antigens of human esophageal carcinoma.When the antibodies were diluted as grear as 1∶2000 and the nuclearmatrix antigens were used in 5 ug/100ul,the content resuIts can be obtained.Our data illustrated that this ELISAfor nuclear matrix antigens is much more sensitive than the indirect immunoflouescence microscope,it was simple,relatively rapid,very reproducible and used easily obtainable and storable reagents.The increased sensitivity is especially usefol for the detection of antibody production in small amounts of sera or culture media,and the analvsis of monoclonal antibodies.
