摘要
目的:建立吗啡成隐大鼠模型,并在此基础上观察伏隔核(ACB)和束旁核(PF)神经元自发放电的变化。方法:按剂量逐日递增原则,l日3次皮下注射1%盐酸吗啡注射液,连续5天,建立吗啡成瘾大鼠模型。用细胞外引导并记录神经元放电的方法,观察ACB和PF神经元自发放电的变化.结果:实验组大鼠与对照组大鼠行为比较有显著差异(P<005)。ACB和PF神经元自发放电的形式、性质虽无明显差异,但神经元自发放电频率的分布有显著差异(P<0.05).结论:成功地建立了吗啡成瘾大鼠模型,并发现吗啡成瘾大鼠ACB和PF中神经元自发放电频率分布有改变,证明吗啡成瘾可使中枢神经系统中相关神经元发生功能改变.
Objective(To establish the model of morphinistic rat and observe the spontaneous discharges of neurons in nucleus accumbens (ACB) and nucleus parafascicularis (PF). Methods: l% morphine hydrochloride was injected into rat's back subcutaneous tissue 3 times every day for 5 days in order to build up the morphinistic model according to the principle of gradually increasing the dose of morphine. The discharges of the neurons in ACB and PF were studied by using the external cell recording method. Results: The behaviors of the rats of experimental group were significant difference (P<0.05) as compared with those in normal rats of control group. Although the forms and properties of discharges of neurons in rats with morphine addiction were same as normal rats, but there was significant difference in distribution of discharge frequency of neurons (P<0.05). Conclusion: The model of morphinistic rat had been successfully established. The changes of the distribution of discharge frequency of neurons in ACB and PF of morphinistic rats demonstrated that the function of related neurons in the center neural system were changed in rats with morphine addiction.
