摘要
D-核糖发酵液离心洗涤得菌体 ,采用超声波破碎 (在 40 k W下 ,工作 4s,间歇 4s,破碎 90次 ) ,制备无细胞抽提液。取无细胞抽提液 80 μL 及 10 0 μmol/ L 葡萄糖、4μmol/ LNADP、0 .6 mmol/ L Tris- HCl(p H=6 .8)、10 μmol/ L Mn SO4溶液各 1m L,在 37℃下保温 10min,测定 A340 num的值 ,对比 NADPH标准曲线 ,测定 D-葡萄糖脱氢酶的酶含量。根据测定的时间及酶蛋白的含量 ,从而确定
The cells were harvested from centrifuged fermented broth of D ribose,them disreputed by ultrasonic wave.The suspended solution were centrifuged and cell free extraction were prepared.The cell free extraction (80 μL and 100 μmol/L glucose?4 μmol/L NADP +?0?6 mmol/L Tris HCl (pH=6.8)?10 μmol/L MnSO 4 (respective 1 mL) mixed together.The mixed solution was treated at 37 ℃ for 10 minutes.The absorption number (A 340 nm ) was detected by spectrophometer 756.
